No, you have to take the cotton yarn out if that's what you used for the provisional CO. It may help if use use a thinner needle to insert into each stitch as you take the cotton out of them, go stitch by stitch until all of the sts are on the needle. That's what is meant by 'picking up' not the same thing as when you pick up from an edge.
