Do I block right on top of the blocking mat?
Or do I use a sheet or something underneath? I'm all ready to go, got the mats out, they're laid out and washed (we purchased them years ago as fatigue mats, never used them, I suppose it is good to hang on to some things). I soaked the Moondance Wingspan with a little soap and tepid water, rinsed and squeezed out excess moisture with cloths. I have pins and blocking wires, don't know if I'll need these. TYIA for the help

You can block directly on blocking mats.

That being said, if you are using something/mats that was/were NOT designed specifically for knitting, you may want to check that it is color-fast first.
Or, as you suggested place a piece of fabric over the mat to hopefully absorb any color/bleed.
Or, only block darker yarns on the mat so any absorbed color/bleed won't show.

I tested the mat surface for colorfastness by rubbing it with a cloth saturated with alcohol. Lucky for me, no color came off, and I now have eight blocking mats, each measuring 2'X2'
